The following calculations for both procedures (Static and Natation) are included on the Sperm Motility Quality Control Worksheet, which can be downloaded and printed from the Forms and Documents page.
These instructions include reporting percent progressive motility. If your procedure does not call for this, simply ignore the steps for counting progressive or non-progressive sperm.
Static method
1. Count only the non-motile and non-progressive sperm in Section One.
2. Count the total sperm in Section Two.
3. Subtract the number of non-motile sperm from the total to get the number of motile sperm.
# Motile sperm calculated =
# Total sperm observed - # Non-motile sperm observed
4. Subtract the total of non-motile and non-progressive sperm from the total to get the number of progressive motile sperm.
# Progressive Motile sperm calculated =
# Total sperm observed - (# Non-motile sperm observed + # Non-progressive sperm observed)
5. Calculate the percent motile as shown below:
% Motile sperm =
# Motile sperm calculated / # Total sperm observed
6. Calculate the percent progressive motile as shown below:
% Progressive Motile sperm =
# Progressive Motile sperm calculated / # Total sperm observed
Natation method
1. Count all progressive sperm in Section One.
2. Replay and count non-progressive sperm in Section One. (If not reporting percent progressive motile, combine steps 1 and 2.)
3. Replay once more and count non-motile sperm in Section One.
4. Add your counts together to calculate motile sperm and total sperm.
# Total motile sperm calculated =
# Non-progressive sperm observed + # Progressive sperm observed
# Total sperm calculated =
# Non-Motile sperm observed + # Non-progressive sperm observed + # Progressive sperm observed
5. Calculate percent motile as shown below:
% Motile sperm =
# Total motile sperm calculated / # Total sperm calculated
6. Calculate percent progressive motile as shown below:
% Progressive Motile sperm =
# Progressive motile sperm observed / # Total sperm calculated
